2024/09/19 Introduction to ASCEM and Pre-requisites for CryoSPARC

Cryo-electron microscopy (cryo-EM) has become a crucial tool for resolving the structural information of biological macromolecules over the past decade. This technique can receive high-resolution specimen images, such as viruses, membrane proteins, and metabolic enzymes. Moreover, these images can be further analyzed through special software like CryoSPARC, and then a variety of near-atomic 3D structural maps will be reconstructed. When observing the morphology of biological sample by traditional TEM, the image quality is easily affected by the process of fixation and dehydration. As a result, Cryo-EM technical platform has solved the issues by rapidly plunge-freezing hydrated samples in vitreous ice, reducing background noise from staining agents, and preserving macromolecular structure in a near-physiological state. In the future, cryo-EM could play an important role in developing new drug-delivery systems, including vaccines and anti-viral drugs.
