研究方向

在近年研究相關經費補助下,我們於2016年發表關於β2-AR blockade可以有效抑制結直腸癌細胞株增生腫瘤生長。更進一步研究發現β2-AR blockade主要作用於KRAS野生型的結直腸癌細胞。然而在臨床應用上,KRAS的分型對於結直腸癌是重要的。因此在臨床上可事先利用基因檢測方式分析結直腸癌病患中KRAS基因表現型,之後篩選出KRAS野生型的病患,並利用β2-AR blockade進行治療。在β2-AR blocker取得方面,主要作用為降血壓藥物,且相對副作用比化療藥物小。因此,在藥物取得方面也較為方便且價格相對便宜 ,對結直腸癌治療提供了重要方向。

基於此研究,我們衍生出一個有趣,新穎並且重要的主題,我們發現突變形KRAS的活性會活化autophagy而使膜上的 β2-AR內吞,因此脫敏化β2-AR blockade的生長抑制。

我們在抗結直腸癌藥物的研究有不錯的成果。我們首先發現Flavopereirine,a natural b-carboline alkaloid extracted from Geissospermum vellosii (Pao pereira)有效抑制結直腸癌生長之機制。Flavopereirine可以透過P53 dependent機制而有效抑制結直腸癌於in vitro and in vivo生長。

此外,在科技部相關研究經費補助下也協助我們進行neddylation作用於骨質疏鬆症研究。我們經由體外及體內試驗証實低濃度的neddylation抑制劑MLN4924可以有效抑制噬骨細胞形成及發育,並且不會對成骨細胞造成傷害,最終能達成治療骨質疏鬆的效果。這部份結果尚在撰寫中,但已獲得台灣,日本及美國專利。

綜合以上,我們是一個多主題發展的實驗室,舉凡腫瘤發生及治療,血管新生,骨質疏鬆症,間質幹細胞,呼吸器誘導之肺損傷等都是我們有興趣之主題。
