我主要的研究方向是找出可用於診斷跟治療肝臟移植排斥和耐受的分子生物標記。在使用急性排斥和自發耐受模式的一系列實驗肝臟移植研究,我證明了細胞外組蛋白H1對肝移植排斥的影響以及相應的自體抗體對誘導耐受的治療潛力(Transplantation 2004 and 2007; Transplant Immunol 2008; J Immunol 2009; Biochem Biophys Res Commun 2013; Clin Dev Immunol 2013)。根據我目前的研究,我們的國際合作者廣島大學和城西國際大學開發出針對組蛋白H1模擬表位的免疫抑制單株抗體(SSV mAb)。抗組蛋白H1抗體(或SSV mAb)不僅可以改善排斥反應,也可改善自體免疫肝炎(Immunology 2010),敗血症(BioMed Res Int 2015) ,以及第一型過敏反應(PLoS One 2016)。此外,我近期確認了肝臟miR-301a是一個具有排斥特異性的生物標記(OMICS 2017)。透過篩選SSV mAb的標靶細胞表面分子鑑定出GAPDH,並發表GAPDH對敗血症的治療潛力(Sci Rep 2014)。GAPDH的多樣功能透過誘導M2巨噬細胞可終止免疫反應也發表在期刊(Biofactors 2018)。
研究特色
除了移植免疫研究外,我在過去10年也做了關於光治療與維生素D3的研究。我建立了非酒精性脂肪性肝炎(NASH)大鼠模式並證明透過誘導維生素D3,光治療對NASH的治療潛力(J Hepatol 2011)。這個研究是第一次證實了光治療和補充維生素D3在脂肪肝疾病動物模式的治療功效,而這為後續的NASH人體治療試驗奠定了基礎。這個發現得到期刊社論(Geier A. J Hepatol 2011)的關注,進而帶領這位主編的研究小組對歐洲的NASH患者進行維生素D3的臨床試驗。除了光治療提高NASH患者的維生素D3的臨床影響外(POL Nurs Prac Res 2018),我們也證實光治療在結腸炎(J Gastroenterol Hepatol 2014),過敏性支氣管哮喘(Adv Tech Biol Med 2014;2:1.)和食物過敏(投稿修改中)實驗動物模式的潛力。
我們也建立了肝癌(HCC)動物模式並證實了循環外吐小體miR-92b對免疫抑制腫瘤微環境生成的影響,而導致活體肝臟移植(LDLT)後HCC的發生和復發。測量LDLT前/後外吐小體miR-92b可預測移植後HCC復發的風險(Am J Transplant 2019)。
